What is Lipinski Rule of 5?

The Lipinski Rule of Five is a set of guidelines used in drug discovery to evaluate whether a chemical compound is likely to be a good candidate for an orally active drug in humans.

A compound is more likely to be orally bioavailable if it does not violate more than one of the following rules:

• Molecular weight under 500 Dalton
• Partition coefficient (logP) less than 5
• No more than 5 hydrogen bond donors
• No more than 10 hydrogen bond acceptors
